During 1:1 with manager, say "I am looking for growth in career. Is there anything I can do additionally? I want to add more to my skill set. Like some things that a Lead can do". These additional tasks will give a sneak peak into the lead role. If you like them and if you are doing them well, after 2-3 months, ask manager for opportunities ... 

Understand that its not just your desire, there has to be a lead position available as well and your manager has to like your performance in that role if available
