Do you have any other drives with spare space? As a temporary solution you could create an extra data file for tempDB which would then be populated by using a mixture of proportinal fill and round robin algorithms. It's not a long term solution, I'm only suggesting it to ensure you do not run out of space in case you cannot have downtime.
What you need to do is to find the cause of the problem. Although restarting the service will give you headroom as TempDB is recreated upon startup.
Out of curiosity is your TempDB presized? If not you may wish to pre-size it in order to reduce the amount of growing that will occur later. Turning on instant file initialization will reduce the impact of data file growths - there is a security risk associated with this though.
Back to the question of why tempdb has grown so large:

What is your biggest table, could it be due to an index rebuild or a sort operation that has been left in an open transaction?

Are you using row versioning? This could be in the form of DDL triggers or one of the snapshot isolation levels.
If you have transactions left open from quite some time back the version store which is held in TempDB will not clear down. Eventually this will lead to all your space being consumed.

How many temp tables are open?
I would definitely check for open transactions, unless you have some massive table this is likely to be the reason why space has not been cleared.

first make sure..ur the single user...

make sure no transactions are happening...

take a backup of tempdb...

stop the sql server service...

restart it ...

tempdb is created again...with the initial size...

if it doesnt ..restart ur server.
